A Study on the Immune Sera of Vibrio vulnificus, Serogroups



Abstract
For preparation and serogrouping of the 0-antisera, V. vulnificus strains (serogroup 01, 02, 04, and 05) were inoculated into BHI broth(1% NaCI). After overnight incubation at 36C incubator, the cultures were heated at 1000 for 2 hours. the heated organisms were then washed twice with N-saline, after final centrifugation, the packed cells were resuspended in N-saline.
The density of the cell suspension was adjusted to that of the MaFarland No. 4. Two rabbits per serogroup strain were each injected intravenously with 0.5, 1.0, 2.0, and 3.Oml amount of the suspension at intervals of 3 to 4 days.
The animals were bled on 6th day after completion of the course of immunization and serum was separated for titration of 0-agglutinins of V. vulnificus.
For diagnostic purpose of clinical microbiology, this study was performed for the preparation and absorption of 0-antiserum of V. vulnificus. The results are as follows.
0-agglutinin titers of the immunized rabbit sera that injected with four serogroup strains of V. vulnificus were determined from 1 : 640 to 1 : 1, 280 by bacterial agglutination test.
For absorption of the non-specific agglutinin of each 0--serogroup antisera, 6ml of 1 : 8 diluted antiserum absorbed by the packed cells derived from growth of 2 tubes of BHI broth (1% NaCI) 10ml was sufficient for the diagnostic purpose of V. vulnificus, the cultures to be examined should be washed twice.
The O-serogrouping for 24 strains of V. vulnificus isolated from human and seafish are summerized in table 6. of these, 14 strains were grouped by four 0-antisera (anti-0l, anti-02, anti-04, and anti-05) ; of the remainder, three were cross-reacted with two or four antisera and the other seven strains did not react with any of the antisera.
